Aim of the present study was to investigate the cryopreservation of pheasant semen by adopting the freezing pellet procedure (Tselutin et al, 1995) with some modifications. Two drop volumes (50 and 80 microlitres) and two thawing temperatures (50°C and 60°C) were tested on semen diluted 1:3 in Lake’s diluent, cooled for 10 minutes at 5°C, cryoprotected with DMA at 6% final concentration and equilibrated for 5 minutes at 5°C. Thawing was performed using a thermoregulated heating plate. Assessment of sperm mobility was performed by Accudenz methodology according to Froman’s procedure (1997). As expected spermatozoa mobility was strongly affected by the freezing/thawing procedure. Neither drop volume nor thawing temperature did affect sperm mobility. However the interaction drop volume*thawing temperature was significant (p<0.01), showing better results with bigger pellets thawed at lower temperature and the opposite volume-temperature combination. Considering that 80 ul pellets are quicker to produce and its thawing temperature is lower, this drop volume seems to be the most advantageous choice for the freezing pellet procedure in pheasant semen.
