The objective of the study was to evaluate the PK/PD profile of the novel atypical opioid drug tapentadol (TAP) (5 mg/kg b.w.) intravenously injected (IV) in seven male rabbit undergoing castration. Plasma concentrations of TAP were determined using an HPLC-FL methods. Peri-operative pain was evaluated by different physiologic parameters, while post-operative pain according to a numerical rating scale. TAP was rapidly eliminated and its concentrations were detected up to 4 hours after administration. It was effective on both peri- and postoperative pain. The minimum plasma effective concentrations reported for humans were exceeded for at least 4 hours. In conclusion, although further studies need to be carried out, TAP seems to be a prospective analgesic drug useful also in veterinary medicine.
