The title compounds I [A = absent, O, S, SO, SO2, NH, N(H)CO; B = (un)substituted aryl, heteroaryl, heterocycle], capable of efficaciously activating the enzymic complex AMPK, and useful for the prophylaxis and therapeutic treatment of diseases and disorders in particular metabolic disorders such as diabetes and obesity, immune-mediated inflammatory pathologies and tumors, were prepared and/or claimed. E.g., a multi-step synthesis of 3-amino-5-phenylbenzofuran-2-carboxamide, starting from 5-bromo-2-hydroxybenzonitrile and 2-bromoacetamide, was described. Exemplified compounds I were tested for the AMPK activation in the C2C12 cell line (data given). Pharmaceutical composition comprising compound I was disclosed.
