The aim of this study was to suggest a more definite, careful and rapid method than the ones used in the past for amylase determination in soil. The main parameters affecting amylase activity in soil, such as pH, temperature, incubation time, soil:solution ratio, were evaluated by laboratory investigation. A substrate solution of starch (1%) and NaCl (1%) was used. Results indicated that the optimal conditions were: pH 6.5, incubation 24 hours at 50 degreesC, and a soil:solution ratio 1:5. The activity of soil amylase was determined by calculating the glucose produced during enzymatic reaction through glucose test strips.
